We want to obtain FLT3-ITD gene signature. To do so, we transduced CB CD34+ cells with mock or FLT3-ITD vectors and performed RNA sequencing (RNA-Seq). Two Groups: Group1: CB CD34+ cells transduced with mock vector; Group2: CB CD34+ cells transduced with FLT3-ITD vector; Overall design: Expression profiling data in FLT3-ITD+ Cord blood cells were compared to corresponding control mock transduced cells by RNA-seq technology

Construction protocol: Total RNA was extracted using Trizol Reagent (Thermo Fisher Scientific) following the manufacture`s instructions. RNA quality was assessed by spectrophotometry(NanoDrop, ND1000) and gel electrophoresis. To be considered as high-quality, the RNA had to have the result of A260/A280≧1.8, A260/A230≧1.5, no gDNA contamination. RNA integrity was measured by Agilent RNA 6000 Nano Assay (RIN≧6). RNA libraries were prepared for sequencing using standard Illumina protocols
